白细胞介素-6对人冠脉平滑肌细胞妊娠相关血浆蛋白A分泌的影响

摘　　要：目的研究炎症因子白细胞介素-6(interleukin-6,IL-6)对人冠状动脉平滑肌细胞(human coro-nary artery smooth muscle cell,HCASMC)分泌妊娠相关血浆蛋白A(pregnancy-associated plasma protein-A,PAPP-A)的影响。方法一期:培养HCASMC,至第5～7代时,分别加入10μg/L和0μg/L的IL-6,0μg/L组设为对照组,均分别孵育2,4,8,24,36h后,收集细胞培养液上清。二期:采用不同浓度的IL-6(0,5,10,50μg/L)(0μg/L组设为对照组)刺激HCASMC6h后分别收集细胞和细胞培养液上清。应用酶联接免疫吸附剂测定(enzyme-linked immunosorbnent assay,ELISA)方法检测细胞培养液上清内PAPP-A的表达量。结果一期,PAPP-A的表达量在2h时开始增加,4,8,24h和36h组浓度显著高于对照组(4h:P=0.008;8h:P=0.007;24h:P=0.023;36h:P=0.017),并随时间的延长而不断增加;二期,在实验剂量范围内,随IL-6剂量增加,PAPP-A的表达量不断升高,其中10μg/L和50μg/L组均显著高于对照组(10μg/L:P=0.015;50μg/L:P<0.001)。结论IL-6可使HCASMC表达不稳定斑块标记物PAPP-A的升高,说明炎症可能是急性冠状动脉综合征(acutecoronary syndromes,ACS)发生发展的机制之一。Objective To observe the effect of IL - 6 on secretions of PAPP - A in HCASMC. Methods HCASMC was incubated with IL -6 （10 μg/L and 0 μg/L）. Culture media was collected after 2, 4, 8, 24 h and 36 h respectively. HCASMC was stimulated with IL- 6（0, 5, 10, 50 μg/L） respectively and culture media was correlated after 6 h. The concentration of PAPP - A in culture media was measured by ELLISA. Results Incubation of HCASMC with 10 μg/L IL -6 resulted in a time - dependent in PAPP - A secretion. It was up - regulation at 2h,and the concentration at 4, 8,24 h and 36 h was significantly higher than that of control group （4 h ： P = 0. 008 ; 8 h ： P = 0. 007 ; 24 h ： P = 0. 023 ;36 h：P = 0. 017 ）. Incubation of HCASMC with different concentrations of IL -6 caused a dose - dependent increase in PAPP - A secreation, and the concentrations of the 10 μg/L and 50 μg/L group were significantly higher than that of the control group（10 μg/L：P =0. 015;50 μg/L：P 〈0. 001）. Conclusion IL -6 can induce the HCASMC＇S production of PAPP - A, suggesting that inflammation may paly an important role in the development of ACS.
