摘  要:目的寻求高效抗血吸虫感染的双价核酸疫苗。方法以共表达和融合表达两种方式构建两种日本血吸虫双价核酸疫苗pVIVO2-mcs-Sj23/SjFABP、pVIVO2-mcs-Sj23·SjFABP,以鸡尾酒式混合获得混合双价疫苗(pVIVO2-mcs-Sj23/SjFABP与pVIVO2-mcs-Sj23·SjFABP)。大量制备质粒,50只BALB/c小鼠随机分为5组:混合疫苗组、双价共表达疫苗pVIVO2-mcs-Sj23/SjFABP组、双价融合表达疫苗pVIVO2-mcs-Sj23·SjFABP组、空质粒对照组及生理盐水对照组。免疫后4周攻击感染,感染后45天剖杀,计数各组小鼠成虫数、肝虫卵数和免疫保力超过50%实验组的虫卵肉芽肿面积。结果与对照组比较,实验组小鼠成虫数、虫卯数和虫卵肉芽肿面积有显著性差异(P<0.05);融合表达组和共表达组无显著性差异(P>0.05);混合DNA疫苗组免疫保护力优于单一双价组;与混合DNA疫苗组比较,pVIVO2-mcs-Sj23·SjFABP组的肝脏肉芽肿面积有显著性减小(P<0.05)。结论血吸虫混合双价DNA疫苗诱导小鼠对血吸虫的保护力优于单一双价DNA疫苗。Objective To explore the high performance bivalent DNA vaccines of Schistosomajaponicum. Methods Three bivalent DNA vaccines were constructed in the methods of the co-expression of two genes, a fusion gene expression and two kinds of plasmids in combination (cocktail vaccine). Large of plasmids DNA was prepared. 50 BALB/c mice were randomly divided into five equal groups: cocktail group、pVIVO2-mcs-Sj23/SjFABP、pVIVO2-mcs-Sj23·SjFABP、pVIVO2-mcs and normal saline group. At week 4 after immunization, each mouse was challenged. At day 45 after challenge the mice were sacrificed. The number of adult worms and hepatic eggs were counted. The sizes of granuloma areas in liver of which protection was higher than 50% were measured. Results The reduction rates of worm/egg/granuloma area of the three bivalent DNA vaccine groups were all higher than that of pVIVO2-mcs group significantly. There appears to be no difference between two genes co-expressed and fusion gene expression in protection. The reduction rate of worm in cocktail group was higher than any single bivalent group. However, the reduction rates of granuloma area in pVIVO2-mcs-Sj23·SjFABP group was higher than cocktail group significantly. Conclusion It was suggested that the protection of cocktail bivalent DNA vaccines was higher than bivalent DNA vaccine.
