通用引物PCR法在人乳头瘤病毒基因检测中的应用

摘　　要：目的建立通用引物PCR法检测人乳头瘤病毒(HPV)基因的方法,探讨其在临床辅助诊断中的应用。方法收集787份标本,其中来自男性160份,女性标本627份。标本均使用通用引物对MY11/09扩增HPVL1基因片段。同时扩增β球蛋白(β-globin)基因作为内对照。结果787份标本中,191份标本HPVL1基因阳性,阳性率为24.3%。男性20～岁组HPV的检出率最高,女性30～岁组HPV的检出率最高,但均无年龄上的显著性差异。患有性病(淋病、尖锐湿疣)、不孕症、宫颈炎、非淋菌性阴道炎等疾病的女性,HPVL1基因的检出率分别为83.3%、20.9%、22.7%、28.0%。男性性病(淋病、尖锐湿疣)、前列腺炎、非淋菌性尿道炎、不育症患者的HPVL1基因检出率分别为42.9%、18.9%、12.5%、7.8%。结论HPVL1基因的检出率较高。通用引物PCR法在临床检验中具有较高的敏感性和特异性。Objective To study the advantages of the MY11/09 PCR in HPV L1 gene detection and the application in the clinic. Methods 787 samples including 160 male and 627 female samples were collected HPV L1 and β-globin genes of all the samples were amplified simultaneously. Results 191 samples out of 787 were HPV L1 gene positive and the positive rate was 24.3%. The highest male HPV L1 gene positive rate was in the age group of 20-,and the female highest HPV L1 gene positive rate was in the age group of 30-,but the differences were not significant. HPV L1 has been detected from patients suffering from some genital diseases, such as colpitis, urethritis, prostatitis, sterility, cervical and cutaneous warts. Conclusions HPV infection is related to some genital diseases. General primer MY11/09 PCR is sensitive and specific for detection of HPV L1 gene in the clinic.
