摘 要:目的观察在急性肝损伤小鼠体内是否存在骨髓来源肝细胞。方法使用CCL4诱导小鼠急性肝损伤,24 h内移植入绿色荧光蛋白转基因小鼠骨髓细胞,移植后不同时间采用免疫荧光方法检测肝脏中的骨髓来源肝细胞。结果骨髓移植后在8周的观察期内肝脏中始终存在骨髓来源的造血干细胞,移植后1、2、4、6、8周肝脏中存在现骨髓来源的表达AFP的未成熟肝细胞,移植后4、6、8周时肝脏中检测到骨髓来源表达ALB的成熟肝细胞。结论急性肝损伤小鼠体内存在骨髓来源肝细胞,骨髓可能是急性肝损伤后肝再生途径之一。Objective To investigate the bone marrow-derived hepatocytes in mice after acute liver injury. Methods After acute liver injury induced by carbon tetrachloroid and transplantation of bone marrow cells isolated from green fuorescence protein (GFP) transgenic mice within 24 h ,murine liver tissues were collected and detected for bone marrow-derived hepatocytes by double immunofluorescence stai- ning. Results Hematopoietic stem cells derived from transplanted bone marrow cells existed in liver sinusold during 8 weeks. Immature hepatocytes expressing AFP differentiated from transplanted bone marrow cells were identified at 1,2 ,4,6 and 8 week after transplantation, while mature hepatocytes differentiated from transplanted bone marrow cells were observed at 4,6,8 week after transplantation. Conclusion Bone marrow-derived hepatocytes can be identified after acute liver injury, and bone marrow may be one of the potential compartments for liver regeneration.
