摘  要:目的观察热量限制对人二倍体成纤维细胞IMR-90衰老相关基因表达的影响。方法应用RT-PCR和Western blot的方法,对含低浓度(3.0mmol/L)、高浓度(22.5mmol/L)葡萄糖和正常对照组(含5.0mmol/L葡萄糖)培养条件下的IMR-90细胞衰老相关基因p16、p21和p53的mRNA和蛋白表达水平进行分析。结果与对照组和高糖培养条件下的早期和晚期细胞相比较,低浓度葡萄糖培养条件下的早期和晚期细胞p16和p21在mRNA水平均显著下降(P〈0.05);低浓度葡萄糖组、对照组和高浓度葡萄糖培养组晚期细胞p16蛋白水平分别为4.2、7.4、9.7,比各自早期细胞表达(0.6、1.0、1.8)增加5~7倍;低浓度葡萄糖组,对照组和高浓度葡萄糖培养早期细胞和低糖培养晚期细胞p21蛋白水平差异无统计学意义,分别为1.1、1.0、1.3、0.9,但与对照组和高糖培养晚期细胞p21蛋白水平(3.1、4.2)相比较则降低3~4倍;各组培养条件下的早期和晚期细胞中p53表达差异均无统计学意义(P〉0.05)。结论各组晚期细胞比各自早期细胞p16基因表达增高;对照组和高糖培养条件下的晚期细胞比各自早期细胞p21基因表达增高;低浓度葡萄糖培养延缓IMR-90细胞衰老并伴有p16和p21基因表达下调;p53基因可能不是IMR-90细胞复制衰老主要调节因子。Objective To observe the effect of caloric restriction (CR) on expressions of senescence-associated genes of IMR-90. Methods Techniques of reverse transcription polymerase chain reaction (RT-PCR) and Western blot were used to assess p16, p21 and p53 genes expressions of IMR-90 cells cultured with low glucose (LG) (3.0 mmol/L glucose), normal glucose (NG) (5.0 mmol/L glucose) and high glucose (HG) (22. 5 mmol/L glucose) DMEM, respectively. Results In early and late cell stage, the levels of mRNA of p16 and p21 of IMR-90 cells were significantly lower in LG group than in NG and HG groups (P〈0.05). The levels of p16 protein were increased to 5-7 folds in the cells in late stage of LG(4.2), NG(7.4) and HG(9.7)groups, when compared with those in early stage of LG(0. 6), NG(1.0)and HG(1.8) groups. Although there were no differences in the levels of p21 protein among early stage of all groups [LG (1.1),NG(1.0)and HG(1.3)] and late stage of LG group(0. 9), and were decreased to 3-4 fold when compared with the cells in late stage of NG(3.1)and HG(4.2) groups. No significant differences were found in the levels of p53 gene expression in all stage among all groups(all P〉0. 05). Conclusions These results suggest that effect of CR on retarding replicative senescence of IMR-90 cells might be associated with low expressions of p16 and p21 but not p53.
