摘 要:以菊花品种‘铺地金’和‘七月桃花’的幼嫩茎段为外植体,从基部至茎尖依次横向切成0.3~0.5 mm的薄层,放在添加不同浓度6-BA、NAA的MS培养基上,进行不定芽的诱导,研究不同激素组合和不同薄层部位对不定芽再生的影响,分别以蔗糖和果糖为碳源,考察碳源种类和浓度对玻璃化的影响。结果表明,适宜于‘铺地金’诱导不定芽再生的最佳培养基为MS+2 mg/L BA+0.5 mg/L NAA,再生率达到96.4%,适合于‘七月桃花’诱导不定芽再生的培养基为MS+2 mg/L BA+0.1 mg/L NAA时,不定芽再生率可高达97.9%,适当提高蔗糖浓度可以有效减少薄层组培中的玻璃化现象,而高浓度的果糖会对外植体产生毒害作用。Using the young stem of chrysanthemum Pudijin and Qiyuetaohua as explants, the stems were cut into 0.3-0.5 mm thin cell layers and put on MS medium with 6-BA and NAA, the effects of different hormone concentration on shoot regeneration were studied. Sucrose and fructose were used as carbohydrate to study the effect of carbohydrate concentration on vitrification. The results showed that the optimum medium of shoot regeneration was MS+ mg/L BA+0.5 mg/L NAA for Pudijin and MS+2 mg/L BA+0.1 mg/L NAA for Qiyuetaohua, the differentiation rate were 96.4% and 97.9% respectively. The vitrification can be effectively reduced by increasing the sucrose concentration and high concentration of fructose was toxic to plants.
