天癸更年软胶囊的抗氧化作用及其肝细胞线粒体膜分子机制

摘　　要：目的观察天癸更年软胶囊所含内容物沙棘脂肪酸对老年大鼠抗氧化作用及其对肝脏线粒体保护作用,探讨其可能的线粒体膜分子机制。方法沙棘脂肪酸给实验组大鼠灌胃45 d,观察沙棘脂肪酸对大鼠血清及脑组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、脑组织单胺氧化酶(MAO)、线粒体MDA和ROS以及脑脂褐素的沉积作用,并观察其对线粒体瞬时通透性孔道(MPTP)的敏感性、线粒体Ca2+含量、线粒体活性氧生成和MDA含量以及电镜下肝脏线粒体超微结构的影响。结果沙棘脂肪酸能显著改善大鼠血清及脑组织和线粒体中多项指标的水平,减轻脑脂褐素沉积及肝脏线粒体损伤。结论沙棘脂肪酸具有抗氧化的能力,可能是通过减少线粒体ROS的产生和脂质过氧化,同时减轻线粒体内钙离子超载,使得老年大鼠肝脏线粒体渗透性非特异孔道的敏感性降低,从而保护线粒体膜及其功能。Objective To observe the antioxidant effect of Tiangui menopause soft capsule contained hippophae rhamnoides fatty acid and its protect effect on hver cell mitochondria in old rats, investigate the probable molecular mechanisms of mitochondrial membrane. Methods Hippophae rhamnoides fatty acid was given by intragastric administration to rats for 45 days to observe its effect on activity of SOD, GSH-Px, MDA in serum and brain, MAO in brain and MDA, ROS in mitochondria, and deposition of lipofuscin in brain. At the same time, its effect on the sensitivity of mitocbondria permeability transition pole （MPTP）, Ca^2＋ contents, active oxygen production and MDA contents in mitochondria and liver mitochondria ultrastructure. Results Hippophae rhamnoides fatty acid obviously improved the levels of some indexes in serum, brain and mitochondria, lightened lipofuscin accumulation in cerebral cortex and damage in liver mitochondria. Conclusions Hippophae rhamnoides fatty acid has antioxidant and antiaging effect, mechanism of which is perhaps related to its effect of decreasing ROS and mitochondrial lipid peroxidation, reducing calcium overload in mitochondria, and decreasing sensitization of MPTP, so as to protect mitochondria membrane and its function and delay the aging process.
