摘 要:目的:观察湿迪胶囊对大鼠佐剂性关节炎(AA)的作用并初步探讨其作用机制。方法:SD大鼠,雄性,随机分为正常组(0.5%CMC-Na溶液)、模型组(0.5%CMC-Na溶液)、地塞米松组(0.45 mg·kg-1)、白芍总苷组(150 mg·kg-1)以及湿迪胶囊(以生药计)高(5400 mg·kg-1)、中(2700 mg·kg-1)、低(1350 mg·kg-1)剂量组;除正常组外,其余各组采用大鼠右后足跖皮内注射氟氏完全佐剂建立AA模型,并分别给予灌胃治疗,观察各组的足跖肿胀度,测定炎性肿胀足前列腺素E2(PGE2)含量及血清中丙二醛(MDA)、超氧化物歧化酶(SOD)含量,测定脾及胸腺指数,并观察踝关节的病理组织学变化。结果:湿迪胶囊高、中、低剂量组可剂量依赖性显著抑制大鼠右后足跖的原发性肿胀和左后足跖的继发性肿胀;与模型组比较,湿迪胶囊高、中、低剂量组SOD总活力明显升高,MDA含量明显下降,PGE2含量降低;与模型组比较湿迪3个剂量组的胸腺和脾脏指数均有所提高,差异有显著意义;湿迪高、中、低剂量组均可明显减轻AA大鼠的病理变化。结论:湿迪胶囊能明显抑制滑膜组织的炎症,减轻细胞的损伤程度,因而对AA大鼠具有一定的治疗效果。Objective: To probe into the effects and mechanisms of Shidi capsule on adjuvant arthritis model (AA). Method: SD rats, maleness, were stochastically divided into the normal comparison, the model, the dexamethasone (0. 45 mg·kg^-1 ), total glucosides of paeony ( 150 mg·kg^-1) and Shidi capsule (gruffs) high (5 400 mg·kg^-1), middle (2 700 mg·kg^-1 ) and low ( 1 350 mg·kg^-1 ) dose groups. Except the normal comparison group, rats in the other groups were injected Freund' s complete adjuvant reagent into the rat' right metapedes to establish the AA model. Drugs were given by intragastric administration. Then paw swelling, the content of prostaglandin E2 in inflamed paws, MDA and SOD in blood serum, the spleen and thoracic gland index, and histopathology change of malleolus joint were observed. Result: The Shidi high, middle and low dose groups could inhibit the primary and secondary swelling of the rats' inflamed metapedes in a dose-dependent manner. Compared with the model group, each group of Shidi capsule could increase SOD,decrease MDA and PGE2, advance the spleen, and thoracic gland index, and relive the histopathology change of malleolus joint. Conclusion: Shidi capsule could inhibit the inflammation of synovial membrane obviously, relieve the degree of injury of cell, and therefore have therapeutic efficacy to AA rats.
