摘  要:目的制备含银脱细胞异种真皮,观察其抑菌效果,为临床上治疗大面积深度烧伤或严重创伤后皮肤损毁提供一种良好的真皮替代物。方法用0.25%胰蛋白酶和0.5%曲拉通(TritonX-100)溶液联合脱细胞法制备脱细胞异种真皮后,用0.2%硝酸银溶液浸泡制备出含银脱细胞异种真皮。以原子吸收光谱仪测量含银脱细胞异种真皮的Ag+含量,并对其进行微生物检测和组织学观察。结果①脱细胞异种真皮基质中的表皮已经完全除去,胶原纤维粗细均匀,排列规则,无明显的变性,真皮中无细胞及细胞碎片存在;②含银脱细胞异种真皮的含银量高,体外抑菌作用明显强于单纯脱细胞异种真皮(P<0.05)。结论含银脱细胞异种真皮在脱除了基质中有免疫原性的细胞成分的同时,又保留了组织的基本结构和完整的胶原纤维支架,从而成为低抗原性的真皮支架,且又因含有银离子而具有较好局部抗菌作用,因而是一种良好的真皮替代物。Objective To prepare xenogenic acellular dermal matrix(xeno-ADM)incorporated with silver and observe the antigenicity and bacteriostasis effect of the xeno-ADM incorporated with silver in order to explore a good idea in the treatment of full-thickness skin in juries or serious wound. Methods Using 0.25 % Trypsin and 0.5% TritonX-100 to prepare xeno-ADM, After that the xeno-ADM was treated with 0.2 % Silver nitrate to prepare xeno-ADM incorporated with silver. Using the Atomic Absorption Spectrometer measures Ag^+ content of the xeno-ADM. The xeno-ADM incorporated with Silver was examined by microbial examination ()and examined microscopically by optical electronic microscope and scanning electron microscopic technique. Results (1) observed under light microscopy, the epidermis was completely removed, there were not any cellular components, hair, follies, sebaceous glands and vessels in the dermis to be found. (2)the Ag^+ content of xeno-ADM incorporated with silver was high. The bacteriostasis test was obviously higher than other groups(P〈0.05). Conclusion The nogenic acellular dermal incorporated with silver is an ideal dermal substitute ,which removed completely the epidermis and cellular components in dermis and retained the integrity structure of collagen fibers, and which have partial, good and strong bacteriostatic effect.
