摘 要:目的:探讨DEHP对体外培养的小鼠附睾上皮细胞生长、活性及功能的影响。方法:将性未成熟期(PND20)和性成熟期(PND50)健康雄性KM小鼠附睾上皮细胞分离后进行体外培养,分别随机分为正常对照组、睾酮组和DEHP组。观察各期各组小鼠附睾上皮细胞的生长增殖情况,MTT法测定附睾上皮细胞活性,取细胞培养液上清测定附睾的功能性指标:唾液酸(SA)含量、α-1、4-糖苷酶活性及乳酸脱氢酶(LDH)活力。结果:DEHP组附睾上皮细胞活性降低,细胞培养液中唾液酸(SA)含量升高、α-1、4-糖苷酶活性及乳酸脱氢酶(LDH)活力降低;正常对照组、睾酮组细胞生长增殖正常。结论:DEHP可抑制体外培养的小鼠附睾上皮细胞生长增殖、活性,导致附睾功能受损。Objective:To study the effects of DEHP on epididymal epithelial cells's growth, viability and function. Methods: Epididymal epithelial cells of PND20 and PNDS0 healthy male Kid mice were separated to culture in vitro, they were randomly divided into normal control group, testosterone group and DEHP group. In each stage and group, growth and proliferation of epididymal epithelial cells were observed by inverted microscope, viability of epididymal epithelial cells were measured using MTT assay, content of Sialic acid(SA) was detected and the activity of α-1,4-glucosidase and lactic acid dehydrogenase(LDH) were investigated from the supematant of medium. Results :Cells grew and proliferated were well in normal control group and in testosterone group, but were inhibited in DEHP group. It showed that viability of epididymal epithelial cells decreased in DEHP group Using MTT assay. The results of epididymal functional index detection showed that activities of the α-1,4-glucosidase and LDH decreased but contents of SA increased in DEHP group. Conclusions: DEHP could induce inhibition of growth and viability of epididymal epithelial cells, and could induce lesions of epididymal function.
