摘 要:用两株流行性出血热病毒(EHF)感染体外培养的人脐带静脉血管内皮细胞(HUVEC),观察EHFV对HUVEC形态学和分泌前列环素(PGI2)的影响。结果发现:感染EHFV的HUVEC未出现明显的细胞病变,但PGI2的分泌显著增加。研究结果揭示:EHFV可直接刺激血管内皮细胞分泌PGI2,可能是引起流行性出血热患者病程初期血浆PGI2水平升高的重要原因。PURPOSE To investigate whether epidemic hemorrhagic fever virus (EHFV) was able to stimulate the PGI2 secretion by vascular endothelial cells in vitro.METHODS Vascular endothelial cells were isolated from veins of freshly acquired umbilical cords, main-tained with TC199 medium, and studied in passages 2 through 5. Endothelial cell cultures were inoculated with two strains of EHFV and examined daily for cytopathic effects. The 6-Keto-PGF1a level in medium of endothelial cell cultures was measured by RIA 3 days after inoculation.RESULTS At no time during the course of EHFV infection was any cytopathic effect observed, the 6-Keto-PGF1a levels in medium in both EHFV infected groups were significantly higher than that in control group.CONCLUSIONS EHFV may stimulate PGI2 secretion by vascular endothelial cells, which may be respon-sible for the elevated plasma PGI2 level in patients with EHF at the early stage of the disease.
