复方黄黛片含药兔血清对NB_4细胞株作用的研究

摘　　要：目的研究复方黄黛片含药兔血清对NB_4细胞株的作用。方法以免为实验血清的供体,以给药与否及血清灭活与否为分组依据,采用双道原子荧光法检测血清中砷的浓度,以细胞抑制率及凋亡率为观察指标,对实验结果进行分析。结果(1)给药前、后兔血清中砷剂浓度分别为(0.01±0.001)mg/L和(0.11±0.006)mg/L,差异显著;(2)四组兔血清对NB_4细胞株生长均有抑制作用,且对NB_4细胞株生长的抑制作用具有一定的浓度及时间依赖性;(3)四组兔血清均可诱导NB_4细胞株发生凋亡,且该作用具有一定的浓度及时间依赖性。结论(1)复方黄黛片含药兔血清可明显抑制人急性早幼粒细胞白血病NB_4细胞株的生长、抑制作用呈一定的浓度、时间依赖性;(2)复方黄黛片含药兔血清可诱导人急性早幼粒细胞白血病NB_4细胞株发生凋亡,该作用呈一定的浓度、时间依赖性;(3)中药血清药理学方法用于深入研究复方黄黛片抗白血病作用机理,能够较真实反映机体血药浓度下的作用及变化规律,能够全面展示复方黄黛片各组分的共同作用,具有可行性和科学性。Objective： To explore the effects of leporid blood serum with compound realgar natural indigo tablet （CRNIT） on cell line NB4. Methods： We took the experimental rabbits as the provider of the animal blood serum. The blood serum was divided into four groups based on whether the rabbits were given CRNIT and whether the blood serum was inactivated. AFS -230a double path atom fluorescence photometer was used to detect the concentration of arsenic in the rabbits blood serum. The inhibition ratios and apoptosis ratios were the observational index. Results： （1）The concentration of arsenic in the rabbits blood serum with and without the medicine were 0.01 ± 0. 001 mg/L and 0. 11 ± 0. 0064 mg/L respectively. （2）The four groups of serum all had the inhibitory effect on the growth of NB4 cells which depended on the medicine concentration and effect time. And there was significant difference among the groups. （3） The four grou of serum all can induce the apoptosis of NB4 with positive relations of the concentration and effect time. And there was significant difference among the groups. Conclusion： （ 1 ） The rabbits blood serum with CRNIT can obviously restrain the growth of NB4 cells and the inhibitory effect depends on the concentration and effect time. （2）The rabbits blood serum with CRNIT can induce the apoptosis of NB4 cell line and the apoptosis ratios depend on the concentration and effect time. （3） The method of Chinese medicine serum pharmacology can relatively truly reflect the effect of medicine in the body, can indicate the collective function of the four components in CRNIT in the round and it is feasible to lucubrate the mechanism of the effect of CRNIT on leukaemia.
