摘 要:目的探讨精子亚临床损伤实验室敏感指标。方法应用计算机辅助精子分析(CASA)、精子染色质结构分析(SCSA)、精子单细胞凝胶电泳分析(SCGE)进行精子测定,比较各项指标在低强度非致热微波辐射和低频震动两种不同暴露环境下对男性精子亚临床损伤的敏感性。根据暴露组与对照组结果之间差异显著性(P值)大小将检测指标分为Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ级。结果精子平均移动角度是Ⅱ级指标;D级精子率是Ⅲ级;A级精子率、精子侧摆幅度、精子鞭打频率、FSH、COMPαt、Ⅰ级慧星率、精子密度是Ⅳ级,精子直线速度、平均路径速度、C级精子为Ⅴ级。结论CASA检测的精子平均移动角度可灵敏反映两种暴露因素对精子的影响,其次是D级精子率。Ⅳ级、Ⅴ级指标是单一暴露因素引起的差异,如COMPat或1级彗星率,可能是造成Ⅰ级、Ⅱ级、Ⅲ级差异的原因,因为精子核DNA损伤,直接影响精子头的运动活力使D级精子率、MAD、VSL、VAP改变。Objective To search the sensitivity of detecting manner on evaluation of the sub-clinical injury of the male reproductive ability. Methods Sperm were detected by computer assisted sperm analysis (CASA), sperm chromatin structure assay (SCSA), sperm single cell gel electrophoresis (SCGE).The hormone level in blood were detected by radiation immunoassay. Compared the sensitivity of each parameter to sub-clinical injury of male reproduction ability in the low intensity nonheating microwave radiation group and the low frequency shaking group. According to significance of difference (P value) of exposed and control group, the parameters were classified into four kinds( Ⅰ , Ⅱ, Ⅲ,Ⅳ , Ⅴ grades). Results The MAD was Ⅱ grade and the rate of D grade sperm was Ⅲ grade. The rate of A grade sperm, ALH,BCF, FSH in blood, COMP α t, the rate of I grade sperm comet and the sperm density were IV grade. The VSL,VAP and the rate of C grade were V grade. Conclusion The MAD and the rate of D grade sperm in CASA were sensitive to the influence of the two exposed factors on sperm. Though IV and V grades were sensitive to one of two exposed factors, but they were the cause of the greater sensitivity of Ⅰ , Ⅱ, Ⅲ grades. For example, when COMP α t and the rate of 1 grade sperm comet increased, the nucleus DNA of sperm was injured greater, that would directly influence the ability of sperm movement and make MAD, the rate of D grade sperm, MAD, VSL and VAP change.
