摘 要:采用MTT法测定了热应激后小鼠脾淋巴细胞增殖的情况,以此作为醇提、碱提、酸醇提、酸提4种提取方法所得黄柏生物碱的筛选依据;采用ELISA法测定了醇提黄柏生物碱对热应激后小鼠脾淋巴细胞IFN-γI、L-4分泌水平的动态影响。结果表明,4种提取方法所得的黄柏生物碱均能促进小鼠脾淋巴细胞的增殖,其中黄柏醇提生物碱对淋巴细胞增殖能力的促进作用最强;热应激使IFN-γ的分泌水平上升I、L-4的分泌水平下降;黄柏生物碱下调IFN-γ的分泌量、上调IL-4的分泌量,使二者的含量均恢复到正常水平。结果证明,黄柏生物碱可显著调节热应激所致的IFN-γI、L-4分泌水平的紊乱。The effects of Cortex phellodendri alkaloid(CPA) on splenic lymphocyte proliferation(SLP) after heat stress were detected by MTT chromatometry. On the basis of the above detection,CPAs extracted by limewater, ethanol, sulfuric acid water and the mixture of sulfuric acid-and-ethanol were choosen. The effects of the optimal CPA on IFN-γ and IL-4 production in mice splenic lymphocytes after heat stress were detected by ELISA. In result, all CPAs extracted by four methods could significantly promote SLP, but CPA extracted with ethanol was the best. The heat stress could stimulate the production of IFN-γ and suppress the production of IL-4. CPA could down-regulate the secretion of IFN-γ and up-regulate the production of IL-4. The result showed that CPA could adjust dramatically the disorder of levels of IFN-γ and IL-4 induced by heat stress.
