摘  要:目的探讨地黄饮子对AD模型大鼠的学习记忆及炎性反应的影响。方法模型组以D-半乳糖腹腔注射合并Aβ注射海马制备AD动物模型,各治疗组分别在造模的同时灌胃地黄饮子和抗脑衰胶囊、哈伯因。运用MORRIS水迷宫检测各组大鼠的学习记忆能力及采用免疫组化方法观察各组大鼠海马神经元TNF-α,IL-1α蛋白的变化。结果治疗组能缩短潜伏期、增加过台次数,第四象限与其它象限游泳距离之间比较也有差异,说明地黄饮子能改善痴呆大鼠空间探索记忆能力,低剂量组已经显效,高剂量组效果更为明显。地黄饮子治疗组IL-1α,TNF-α含量显著下降,且具有剂量差别。结论地黄饮子对痴呆模型大鼠学习记忆空间探索能力有改善作用及能有效抑制海马神经元炎性反应,其作用机理可能与地黄饮子抑制海马神经元TNF-α,IL-1α蛋白,抑制小胶质细胞活性进而减轻海马神经元的炎症损伤有关。Objective To investigate effects of Decoction of Rehmanniae(DR) on improving learning memory ability and inflammation reaction of Alzheimer's disease(AD) model rats. Methods AD model rats induced by D- galactose intraperitoneal injection and the hippocampus injected β -amyloid(Aβ) were made. Every group was fed with Hup A, Kangnaoshuai and DR. The ability of learning memory was measured by Morris water maze(MWM). The inflammatory reaction of hippocampus neurons were observed by electron lens using immunohistochemistry method. Results DR could shorten the escape latency and increase the number of passing plats and the swimming distance of the fourth quad compared with the other quads. DR could dose - dependly reduce the expression of TNF-α, IL-1α protein of Hippocampus neurons. Conclusion DR can improve the ability of spatial exploratory and effectively control the inflammation reaction. The mechanism of action is possibly relevant that DR can decrease the expression of IL-1α and TNF-α to inhibit the activity of glia cell and inflammatory injury of hippocamus neurons.
