绵羊肺炎霉形体单克隆抗体的研制及初步应用

摘　　要：用绵羊肺炎霉形体（ＭＯ）抗原免疫ＢＡＬＢ／ｃ小鼠，融合其脾细胞和ＳＰ２／Ｏ骨髓瘤细胞，筛选出６株分泌抗ＭＯ的单克隆抗体（ＭｃＡｂ）杂交瘤细胞系。用间接ＥＬＩＳＡ法对３株ＭｃＡｂ做特异性分析，该ＭｃＡｂ只特异地与ＭＯ抗原发生反应，而不与猪肺炎霉形体、山羊无乳症霉形体抗原发生反应。用检测ＭＯ抗体的间接ＥＬＩＳＡ与ＩＨＡ比较检测绵羊血清中抗ＭＯ抗体，前者的阳性检出率较后者高７．Using Mycoplasma ovipneumoniae (MO) antigen to immunize BALB/c mice,and fusing the aplenocyte with the SP 2/O myelona cell,6 hybridoma cell lines of McAb of secrelory MO have screened.With indirect ELISA specificity of 3 McAb is analysed,the McAb only reacted with MO antigen,but not with Mycoplasma hyopneumoniae,Mycoplasma agalactiac.Compared the indirect ELISA measuring MO antibody with IHA for anti MO antibody level in sera from sheep, the positive detectable rate of former was higher 7.14% than the latter.
