摘 要:通过继代草、日本看麦娘幼穗愈伤组织观察杂草对绿黄隆的诱导抗性特点。结果表明:继代过程中随着绿黄隆浓度逐渐增加,杂草愈伤组织内乙酰乳酸合酶(ALS)对绿黄隆的敏感性(以IC50值表示)明显下降,草愈伤组织经6~12次继代,IC50约从1μg/L增至300μg/L;日本看麦娘愈伤组织经4~10次继代,IC50约从1μg/L增至100μg/L以上。说明杂草愈伤组织对绿黄隆的诱导抗性明显增强,并且草的这种诱导抗性也在再生小植株上表达,表现为200μg/L绿黄隆对抗性再生小植株的生长抑制减弱;1000μg/L绿黄隆对抗性再生小植株的致死率明显变小。而在继代过程中绿黄隆并不显著改变这两种愈伤组织的生长状况。Characteristics of resistance response to chlorsulfuron was observed in young panicle calli of both weeds via subculture. During subculture sensitivity of acetolactate synthase (ALS) in both calli greatly decreased along with increase of chlorsulfuron concentration. IC 50 value (concentration causing 50% inhibition) of ALS activity increased from 1 0 μg/L to 300 μg/L after 6-12 subcultures in Beckmannia syzigachne , or from 1 0 μg/L to over 100 μg/L after 4-10 subcultures in Alopecurus japonicum , indicating that obvious development of the herbicide resistance has appeared in both calli. Moreover, this inductive resistance also in regenerated plantlets of Beckmannia syzigachne . Inhibition of resistant plantlet growth at 200 μg/L of chlorsulfuron lowered, and lethal rate of these plantlets at 1 000 μg/L of chlorsulfuron became smaller. Neither growth of both callus nor calli differentiation potential and plantlet development of Beckmannia syzigachne was influenced by chlorsulfuron during subculture.
