摘 要:目的建立一种重复性好、能快速简便获得高纯度滋养细胞的培养方法。方法取孕6~8周正常绒毛组织40例,分别采用组织贴块法、酶消化法和改良联合法进行原代培养。结果改良联合法培养的细胞生长迅速,接种6~8d即可传代,原代细胞生长时间明显缩短,细胞纯度达90%。结论改良联合法综合了组织贴块法和酶消化法的优点,可快速获得实验所需的人绒毛膜滋养层细胞。Objective To modify a better primary cell culture method of trophoblastic cells. Methods The villus(40 cases) obtained from early pregnancy women (6-8 weeks) were cultured by tissue culture method, enzyme-digestion method and combined method respectively. Results The growth time of primary human trophoblastic cells in combined method group was significantly shorter than that of the other groups. The cell purity was 90 %. Conclusion The combined method with the advantages of tissueculture method and enzyme-digestion method could increase the yields of primary human trophoblastic cells significantly.
