摘 要:中药及其复方发挥疗效具有整体性、多靶点和多成分协同作用的特点。中药药效物质基础的阐明是中药质量控制的基础,是诠释中药作用奥秘的关键。生物活性筛选/化学在线分析技术是由生命科学与色谱、质谱分离鉴定技术交叉形成的一种综合技术,在国内外取得了瞩目的进展,当前已逐渐发展成为中药活性成分筛选和药效物质基础研究的重要手段之一。本文对近年来生物活性筛选/化学在线分析技术的发展做了简要综述,主要介绍了生物色谱法、脂质体平衡透析/液相色谱-质谱联用方法、细胞固相萃取/液相色谱-质谱联用法和微透析生物取样/液相色谱-质谱联用法的原理及其在中药活性成分筛选和药效物质基础研究中的应用。Curative effects of traditional Chinese medicines (TCMs) and combined prescriptions are integrative results of many bioactive components, and TCMs have typical characteristics of synergistic actions and multiple target sites. The screening and analyzing of the bioactive components in TCMs are very important not only for the quality control of the crude herb substances but also for elucidating the therapeutic mechanisms of TCMs. Biological screening-chemical analysis is a interdisciplinary subject which been attracting ever-increasing attention combines the advantages of life sciences and chromatographic techniques. It has and regarded as a powerful tool for the screening of the bioactive components in TCMs. The article reviews the development of biological screening-chemical analysis methods, including bio-chromatography, liposome equilibrium dialysis, cell solid phase extraction, microdialysis sampling coupled with high-performance liquid chromatography or liquid chromatography-mass spectrometry, etc, and introduces in detail their fundamentals and applications in the screening of the bioactive compounds in TCMs.
