摘  要:以多糖提取率为指标,采用单因素实验和正交实验对苦丁茶多糖的提取工艺条件进行优化,确定最佳提取工艺为:水料比20:1、80℃、提取50 min。在此条件下,苦丁茶多糖提取率为2.43%,先后采用Sevag法脱蛋白、分级醇沉法、DEAE—52纤维素离子交换柱层析法对其进行分离纯化,最后得到K_1和K_22个组分。同时对各苦丁茶多糖进行理化性质分析,结合Sevag法预实验所得出的结果,推测苦丁茶多糖可能为以某种方式结合蛋白的糖缀合物。Optimizing the technology of extracting polysaccharides from Ilex kudingcha C. J. Tseng through one--factor experiment and orthogonal design experiment as guiding by the extraction rate of polysaccharides, we determined that the optimum extraction technique is. water to material ratio 20 : 1, extracting temperature 80*(2 ,extracting time 50min. Following the above mentioned conditions, the extraction rate of polysaccharides is 2.43 ~.. By separating the polysaccharides by Sevag way, grade-precipitation with ethanol, DEAE-52 cellulose column chromatograph, we obtained two compositions named K1 and K2. We also analyzed physicochemical properties of overall polysaccharides. According to the anticipated results from Sevag way experiment, we estimated that the polysaccharides from Ilex kudingcha C. J. Tseng are glyco- conjugate combined with protein.
