摘 要:目的观察中华眼镜蛇毒组分H预防性给药对大鼠缺血再灌注心肌细胞凋亡及相关凋亡基因Bcl-2和Bax表达的影响。方法30只SD大鼠随机分为对照组、缺血再灌注组、组分H组各10只。采用原位末端标记(TUNEL)法标记凋亡心肌细胞,免疫组化法测缺血再灌注心肌细胞Bcl-2及Bax的表达。结果心肌TUNEL阳性指数组分H组为(26.2±3.5)%,缺血再灌注组为(34.6±5.3)%,对照组为(3.4±1.8)%,3组比较差异有统计学意义(P<0.01),尤以缺血再灌注组更明显。缺血再灌注组Bcl-2表达降低(0.113±0.02),与对照组(0.146±0.05)比较差异有统计学意义(P<0.01);组分H组Bcl-2表达(0.128±0.03)与缺血再灌注组比较显著提高(P<0.01),但仍低于对照组(P<0.01)。缺血再灌注组Bax表达升高(0.105±0.03),与对照组(0.070±0.01)比较差异有统计学意义(P<0.01);组分H组Bax表达(0.084±0.02)虽比缺血再灌注组降低(P<0.01),但仍高于对照组(P<0.01)。结论中华眼镜蛇毒组分H有显著抗缺血再灌注心肌细胞凋亡作用,调控心肌细胞凋亡基因Bcl-2及Bax蛋白表达可能是其重要机制之一。Objectives To study the effect of prophylactic admistration of fraction H of Naja naja Venom on ischernia-reperfusion rats,and the influence on the expression of cardiomyocyte apoptosis and the gene Bcl-2 and Bax. Methods Thirty adult SD rats were divided into three groups ,fraction H of Naja naja Venom group,ischemia-reperfusion group and control group. The expression of apoptotic cardiomyocyte was determined by Tunel staining. Immunohistochemistry was used to detect mRNA the expression of Bcl-2 and Bax in myocardial cells of rats with ischemia reperfu- sion. Result The apoptotic index was significantly increased in both fraction H of Naja naja Venom group(26.2±3.5)% and ischemia-repeffusion group (34.6±5.3)% (P 〈0. 01) ,especially in fraction H of Naja naja Venom group. The expression of Bcl-2 was decreased significantly in ischemia-reperfusion group than in control group (0. 113±0.02 VS 0. 146 ±0.(15) (P 〈0.01) ,the expression of Bcl-2 in fraction H of Naja naja Venom group was higher than ischemia-reperfusion group, but still lower than control group (P 〈 0. 01 ). The expression of Bax significantly in ischemia-reperfusion group than in control group (0. 105 ±0.03 VS 0.070 ±0. 01) (P 〈0.01 ) ,the expression of Bax in fraction H of Naja naja Venom group was lowerr than ischemia-reperfusion group,but shill lower than control group (P〈0.01). Conclusion Fraction H of Naja naja Venom may effectively protect cardiomyocyte apoptosis induced by ischemia-reperfusion,and one of the mechanisms of it may be adjust the expression of protein Bcl-2 and Bax.
