摘 要:为了能方便快捷地鉴别出铁皮石斛试管苗和铁皮石斛,采用随机扩增多态性DNA(RAPD)技术对铁皮石斛试管苗及石斛属23个种进行了基因组DNA多态性分析,从中找出铁皮石斛试管苗特征性条带进行克隆和测序,并根据测序结果设计了特异性引物.采用此特异性引物对铁皮石斛试管苗及石斛属其他一些种进行扩增,结果表明,铁皮石斛试管苗与各地产野生铁皮石斛出现同一特征条带,而石斛属其他种未出现该条带.To identify the original species of tissue culture seedlings of Denclrobium candidum, randkn amplified polymorphic DNA(RAPD) was used to analyze the genetic relationships of tissue culture seedlings of Dendrobium candidum and other 23 species of Dendrobium. A specific electrophoresis band of tissue culture seedling of D. candidum was found. According to the result of clone and sequence analysis, a spedfic primer was designed and used to identify tissue culture seedlings of D. candidum from other Dendrobium species effectively. Results show that there is a same band in tissue cul- ture seedlings of D. candidum and D. candidum, but not be discovered in other species of Dendrobiun.
