龟鹿二仙胶汤及其拆方对关节软骨细胞增殖的作用

摘　　要：目的:分析龟鹿二仙胶汤方、鹿茸、龟板、人参和枸杞药理血清对大鼠关节软骨细胞的调节作用,探讨中药治疗骨性关节炎的机理。方法:使用酶消化法,建立大鼠关节软骨细胞培养体系;采用MTT法,筛选出龟鹿二仙胶汤、鹿茸、龟板、人参和枸杞作用软骨细胞的最佳药理血清浓度。通过形态学、细胞超微结构观察、MTT法、流式细胞仪细胞增殖指数测定,分析龟鹿二仙胶汤方、鹿茸、龟板、人参和枸杞含药血清以及IGF-I对照组对软骨细胞增殖的影响。结果:建立了大鼠关节软骨细胞培养体系。通过MTT法筛选了龟鹿二仙胶汤及其拆方最佳药效的药理血清。龟鹿二仙胶汤药物血清组、IGF-I对照组能显著促进软骨细胞的增殖,其余各拆方组与正常组比较,无显著性差异,药效低于全方组。结论:证实了药物血清的制备与获得应通过实验筛选得出。龟鹿二仙胶汤能显著促进软骨细胞的增殖,全方药效优于拆方各组,体现了中药复方配伍的优越性。深化了中医"肾主骨"理论与软骨细胞增殖的关系的认识,进一步丰富了中医"肾主骨"理论的现代科学内涵。Objectives： To discuss the effects of medicated serum of GUILUERXIANJIAO formula, LURONG formula, GUIBAN formula,ENSENG formula and GOUJI formula on chondrocyte, to investigate the therapeutic mechanism of Chinese herbs on osteoarthritis, and to developed TCM theory of ＂kidney governs the bone＂. Methods： Chondrocyte culture system in rats was constructed by enzyme digestion method. The best concentration of medicated serum of GUILUERXIANJIAO formula, LURONG formula, GUIBAN formula, RENSENG formula and GOUJI formula was determined by MTT method. The effects of medicated serum of above formula and IGF-I on proliferation of chondrocyte were analyzed by morphological results, ultrastructure observation and cell proliferation index from Flow Cytometry. Results：Chondrocyte culture system in rats was constructed in this research. Through the MTT law screening, we found the best effect medicated serum from GUILUERXIANJIAO formula and its sub--formula. GUILUERXIANJIAO formula and IGF-I could obviously promoted proliferation of chondrocyte. Compared with the normal control group, the sub--formula groups from GUILUERXIANJIAO formula did not show obvious difference and their effects were lower than that of GUILUERXIANJIAO formula. Conclusion：By the production of medicated serum from GUILUERXIANJIAO formula and its sub--formula, we verify that the production of medicated serum from Chinese herb formula should be completed by screening test. GUI- LUERXIANJIAO formula and IGF-I can obviously promoted proliferation of chondrocyte, the curative effect of the whole GUILUERXIANJIAO formula is better than that of all the sub--formula. This phenomena showed the superiority of the Chinese herbs compound prescription. This research can deepen our recognition of the relationship between TCM theory of ＂kidney. governs the bone＂and proliferation of chondrocyte. It further complements the modern scientific concept of TCM theory of ＂kidney governs the bone＂.
