摘  要:参考GenBank中发表PRVOSU毒株VP6序列ORF两端保守区,设计1对特异引物,以PRVJS株反转录cDNA为模板,通过PCR方法获得约1.3kb的基因片段,并克隆到pMD18-T载体中进行序列测定。测序结果表明:获得了VP6的1320bp全基因序列;将其ORF与国内外9株PRVVP6基因的ORF进行核苷酸和氨基酸同源性比较,核苷酸同源性在77.1%~91.1%之间,氨基酸的同源性在89.7%~99.5%之间;在氨基酸系统发育进化树中,JS毒株与A131、OSU、CN86毒株亲缘关系较近.为一个进化群。Based on VP6 sequence of PRV OSU strain published in GenBank, a pair of primer within the conserved region flanked the ORF was designed, and 1.3 kb fragment was amplified by PCR, which was then inserted to the pMD18 -T vector and the sequence was analyzed. The result showed that complete sequence ( 1 320 bp) of JS VP6 gene was obtained. JS VP6 gene has a high sequence homology to those of 9 PRV isolates. The homology of nucleotide sequences and amino acids sequences were 77. 1% - 91.1% and 89.7 % -99.5 %, respectively. JS strains had a closer relationship with A131, OSU, CN86 strains by phylogenetic trees of VP6 of PRV, so they maybe belong to the same group
