摘 要:目的:探讨酸性氧化电位水(EOW)和碱性氧化电位水(AOW)替代硫酸-重铬酸钾液清洗玻璃器皿,用于细胞培养、生物材料、生化检测等实验的可行性。方法:取玻璃培养瓶、试管和吸管各100个(只),洗涤灵洗刷、自来水冲净,烤干后分3组浸入各浸泡液中。A组:硫酸-重铬酸钾洗液;B组:EOW;C组:AOW。浸泡24h后取出,自来水冲洗10遍,双蒸水2遍、三蒸水1遍,烤干,高压蒸汽灭菌后接种RSC-364大鼠滑膜细胞,分别进行细胞生长速度、贴壁率、存活率和活细胞形态比较。结果:细胞生长速度测定:1~3d为对数长生期,平均倍增时间A组为26h,B组34.7h,C组25.8h;4~5d为平台期,细胞生长减慢;第6天细胞停止生长,增殖曲线迅速下降。细胞贴壁率:A组74%,B组65%,C组70%(P>0.05)。细胞存活率:A组93.5%,B组94.1%,C组94.6%(P>0.05)。三组细胞形态变化不大,接种后第1天,95%的细胞由圆形逐渐贴壁成长梭形;第5天细胞长满瓶壁,生长减慢。结论:A、B、C三组细胞生长速度接近,酸、碱性氧化电位水均可替代硫酸洗液清洗玻璃器皿,用于细胞培养。Objective: To study the effect of electrolyzed oxidizing water and alkalized oxidizing water replacing sulfuric acid solution on the cleaning of cell culture glassware. Methods: The glass culture bottles, test tubes and suction tubes were randomly divided into three groups. Group A immersed in sulfuric acid, group B in electrolyzed oxidizing water (EOW), group C in alkalized oxidizing water (AOW), 24 hours later, they were dealt with asusural, and then cultivated RSC- 364 cells. The cell growth rate, attachment efficiency, survival rate and cell shape were compared respectively. Results: Cell doubling time was 26 hours in group A, 34.7 hours in group B, 25.8 hours in group C. Attachment efficiency of group A was 74%, group B was 65% and group C was 70%. Cell survival rate of group A was 93.5%, group B was 94.1% and group C was 94.6%, The changes of cell shape were not obvious. Conclusion: The cell growth rates of three groups are similar. The application of electrolyzed oxidizing water and alkalized oxidizing water has substantial value for replacing sulfuric acid solution on the cleaning of cell culture glassware.
