摘 要:目的:探讨槲皮素防治动物实验性慢性肾功能衰竭时对肾组织病理改变的影响。方法:给实验大鼠饲以含腺嘌呤饲料,饲养2月造成慢性肾功能衰竭;干预组口服槲皮素(100 mg/Kg.d×42 d)治疗;结束时精称存活大鼠体重和肾脏重量并计算肾脏指数;烘干法测定肾脏组织含水量;常规方法对肾脏行病理学检查;测定羟脯氨酸以观察肾组织纤维化程度;免疫组织化学染色法观察肾组织NF-κB变化。结果:与肾衰对照组比较,干预组大鼠肾脏指数和肾组织含水量均显著降低(5.08%±0.89%和78.24%±1.25%vs 12.63%±2.94%和79.29%±0.90%),肾脏病理学损伤改变明显较轻,肾组织纤维化显著减轻(平均每肾羟脯氨酸含量735.5μg±377.7μg vs1540.3μg±358.9μg),肾组织细胞NF-κB P65丢失情况明显减轻。结论:槲皮素对大鼠实验性慢性肾功能衰竭具有显著防治作用,治疗后肾脏病理改变显著减轻。objective: The prevention and curing effects of Quercetin on renal pathological changes in rats with experimental chronic renal failure studied. Methods: The rats in control and treating groups were fed with 0. 75% adenine in feed for 2 month to be build up chronic renal failure, and the quereetin were administered orally ( 100 mg/Kg · d) for42 clays to the rats in treating grouμ beginning at the 15th day. The body and kidney weights of the survived rats were precisely measured and the kidney - indexes were calculated at the end of the treating period. The renal - moisture capacities were determined with a baking method, and the renal pathological inspects were executed routinely. The renal tissue sclerosis was assayed by determining its hydroxypmlfine (Hyp) . The NF - κB in the renal cells were assayed with a immunochemical assay. Results. When compared with the control grouμ significantly ( P 〈 0. 01 or P 〈 0. 001 ), the kidney - indexes and the renal moisture capacities were decreased (5.08% ± 0. 89 & 78.24% ± 1.2.5% vs 12. 63% ± 2. 94 & 79. 29% ± 0. 90% ), the renal pathological changes were less serious, the renal tissue sclerosis changes were reduced ( mean quantity of Hyp per kidney, 735.5 μg ± 377.7 μg vs 1540. 3 μg ± 358.9 μg), and the losses of NF - r B P± in the renal cells were less serious. Conclusion: The results suggested that qercetin is of prevention and curing effects on chronic renal failure in rats, the pathological changes were, evidently reduced after quercetin treatment.
