雪莲注射液抗大鼠佐剂型关节炎和免疫调节作用

摘　　要：目的研究雪莲注射液(SII)的抗大鼠佐剂型关节炎和对免疫功能的影响,为其临床应用提供药理学参考。方法采用大鼠足跖注射Freundi’s完全佐剂形成佐剂型关节炎,应用MTT法检测小鼠T、B淋巴细胞增殖,用羊红细胞免疫法测定小鼠血清溶血素。结果注射佐剂的大鼠右后足跖从第2天开始肿胀,持续至22d(原发性损害);未注射佐剂的左后足跖从第6天开始肿胀,持续至20d(继发性损害);肿胀程度与NS组比较均有明显差异。大鼠im0.2,0.4,0.8mL·kg-1·d-1的SII,22d。对大鼠佐剂型关节炎的原发性和继发性损害都有抑制作用,从第6天开始,明显降低关节的肿胀度(P<0.05或P<0.01)。SII在1∶32～1∶256稀释度范围内对T淋巴细胞增殖有促进作用(P<0.01);在同样浓度范围内对B淋巴细胞增殖则有明显抑制作用(P<0.01)。0.25,0.50,1.00mL·kg-1·d-1的SII,im,8d,可使血清溶血素形成明显减少(P<0.01)。结论雪莲注射液具有抗大鼠佐剂型关节炎和免疫调节作用。Objective To investigate the effect of Saussurea involucrata Injection （SⅡ） on counteracting adjuvant - induced arthritis and its immunoregulation function, thus to supply the pharmacological evidences for the clinical treatment of arthritis. Methods Adjuvant arthritis was induced by plantar injection of Freundi＇s complete adjuvant. MTT method was used to detect T and B lymphocytes proliferation, and sheep red blood cell immunization was used for hemolysin determination. Results （1） Rats right posterior metatarsus which was injected adjuvant was swollen from the second day to the 22nd day （the primary injury）, and left posterior metatarsus not receiving adjuvant injection was swollen from the sixth day to the 20 th day （the secondary lesion）. The differences of swelling degree were significant between SⅡ group and NS group. （2） Consecutive intramuscular injection of SⅡ 0. 2, 0. 4, 0. 8 mL·kg^-1·d^-1 for 22 days suppressed the primary injury and the secondary lesion of adjuvant arthritis in rats, relieved swelling significantly from the sixth day（ P 〈0. 05 or P 〈0. 01 ）. （3） SⅡ promoted the proliferation ofT lymphocyte in the dilution rate of 1：32-1： 256 （ P 〈 0. 01） while inhibited B lymphocyte proliferation significantly in the same concentrations. （4） Consecutive intramuscular injection of SⅡ 1.00 mL·kg^-1·d^-1 for 8 days diminished hemolysin pro- duction significantly（ p 〈0. 01）. Conclusion SⅡ has anti - adjuvant - induced arthritis and immunoregulation functions.
