摘  要:目的建立人工蛹虫草子实体多糖的提取与含量测定方法。方法采用正交试验筛选提取条件,苯酚-硫酸法测定多糖的含量。结果多糖的最佳提取条件为80倍量纯化水,45℃提取1次,每次10 min;线性范围为10~100μg·mL^-1,r=0.9998,加样回收率为99.41%,RSD=2.36%。3批供试品中多糖的平均含量为22.12%。结论该提取和测定方法操作简单,重复性好,结果准确可靠,可用于人工蛹虫草中多糖的提取与含量测定。Objective To develop a method for extracting and determinating polysaccharides from cultured Cordyceps militaris L. Link. Methods Orthogonal experiment was used to confirm the way of extracting. Contents of polysaccharides were detected by using phenol-sulphate acid . Results The best condition for extracting polysaccharides was by 80 times of pure water, once at 45℃, 10 min for one time. Linear range was 10 ~ 100 g · mL^-1 ,r = 0. 999 8, the recovery rate was 99.41% ,RSD = 2.36% o The average concentration of polysaccharides in three samples was 22. 12%. Conclusion The extracting method is simple, reproducible and the results is accurate and reliable. It can be used to extract and deternline the contents of polysaccharides in cultured Cordyceps militaris L. Link.
