摘 要:目的研究短期多不饱和脂肪酸增高对大鼠肝脏TG含量和胰岛素抵抗的影响。方法给大鼠输注脂肪乳,行清醒高胰岛素-正血糖钳夹试验,观察脂肪乳对葡萄糖利用和肝葡萄糖产生的影响。结果脂肪乳使血浆FFA和TG升高,使肝脏TG和肝葡萄糖产生亦明显升高,钳夹状态葡萄糖利用显著降低。结论短期多不饱和脂肪酸增高可引起肝脏TG蓄积和胰岛素抵抗。Objective To investigate the effect of short-term elevation of free fatty acid (FFA) level on hepatic triglyceride(TG) content and insulin resistance. Methods Intralipid plus heparin (IH) were infused into rats for 7 h intravenously. Hyperinsulinemic-euglycemic clamp was performed to examine glucose utilization (GU) and hepatic glucose production (HGP). Results Compared with infusion of saline, infusion of IH elevated plasma FFA, TG, and hepatic TG and HGP. On clamp state, GU was decreased with IH infusions versus saline infusion. Conclusions Short-term elevation of FFA can induce hepatic TG accumulation and insulin resistance. TG plays an important role in development of insulin resistance.
