摘 要:目的:利用基于核磁共振的代谢组学方法探讨雷公藤甲素的肾脏毒性。方法:以生物核磁共振结合模式识别技术和主成分分析法探讨雷公藤甲素口服给药对大鼠尿液内源性代谢产物的影响。结果:给药后,尿样中甘氨酸、醋酸盐、甜菜碱以及丙酮水平显著上升,分别显示首先是肾脏皮层S1受损伤,然后肾乳头受损伤,最后肾脏皮层S3段受到损伤。主成分分析表明,给药组与对照组的代谢谱有明显差异。结论:大鼠尿液的代谢物谱与雷公藤甲素对肾脏造成损害作用密切相关。利用代谢组学方法可以迅速、简便地分析雷公藤甲素肾脏毒性。Objective: To explore the renal toxicity of triptolide with NMR-based metabonomic method. Methods: To analysis the influences of oral dosing of triptolide on endogenetic metabolites in rat urine using bio-nuclear magnetic resonance (NMR) coupled with pattern recognition technique and principal component analysis (PCA). Results: Dosing of triptolide resulted in increases of the concentrations of acetate, betaine and acetone in urine, which suggested the damages on the renal cortex S1, papilla and cortex S3, respectively. PCA revealed that the matabonomic profile of triptolide-treated group was obviously different from that of control one. Conclusion: The metabonomic profile of urine closely related with the renal toxicity of triptolide. Using metabonomic method the renal toxicity of triptolide could be analyzed simply and rapidly.
