摘  要:目的:探讨HHK(人发角蛋白)在桥接周围神经缺损后,诱导神经再生及与周围组织的相容和自身的降解情况。方法:用PLGA(聚乳酸-聚羟基乙酸共聚物)套管装配HHK来桥接SD大鼠10mm长的坐骨神经缺损,术后3、6、9、12周取材,切片做组织学观察。结果:术后3周HHK毛小皮已经开始剥脱降解,并已有大量的雪旺细胞和神经纤维沿HHK长入,6周时HHK已均质化,9周、12周HHK继续降解,长入其间的雪旺细胞和神经纤维更多,排列更加规律。结论:HHK有很好的生物相容性,和可控的降解速率,能很好的诱导神经再生,可以作为支架材料应用于周围神经组织工程研究。Objective: To investigate the compatibility and degradation of human hair keratin (HHK) bridging for peripheral nerve defect and regeneration of the peripheral nerve (PN) induced by HHK, Method: The sciatic nerve of SD rats were transected with 10mm gap, then repaired by the polyactic - polylycolic acid (PLGA) tube with HHK, Histological examination was carried out at 3rd. 6th, 9th and 12nd week after operation, Results: Large amount of schwann cells and nerve fibers were observed under optical microscope to regenerate along HHK, and the hair cuticle of HHK was denudated and degradated at the 3rd week; homogenization of HHK occurred when observed at the 6th week. The degradation of HHK keep on, and more and more Schwarnn cells and nerve fibers with remarkable polarity growed in to the graft at 9th and 12nd week. Conclusion: HHK have controlled degradable rate and good biocompatibility, and can induce the PN regeneration and be used as scaffold material for PN tissue engineering research.
