摘  要:目的:研究CD151对脐静脉内皮细胞(HUVEC)丝/苏氨酸蛋白激酶(Akt)、磷酸化Akt-Ser473(p-Akt-Ser473)表达和细胞增殖的影响。方法:通过构建pAAV-CD151质粒并转染HUVEC,HUVEC分为正常对照组、绿色荧光蛋白(GFP)组和CD151组。Western Blot检测CD151及Akt、p-Akt-Ser473表达,MTT法测定脐静脉内皮细胞的增殖能力。结果:①CD151组CD151及p-Akt-Ser473表达明显增加,CD151组与正常对照组和GFP组比较有显著性差异(P<0.05),而总Akt三组间无明显差异(P>0.05)。②MTT法检测正常对照组、pAAV-GFP转染组、pAAV-CD151转染组的A值分别为0.1663±0.0095、0.169±0.0085和0.2457±0.0044,pAAV-CD151转染组较pAAV-GFP转染组和正常对照组细胞增殖能力增强,且有显著性差异(P<0.05)。结论:CD151具有促进内皮细胞增殖的作用。CD151可能是通过激活p-Akt-Ser473,增加Akt活力,达到促进细胞增殖的作用。Objective:To investigate the the effect of CD151 on Akt-Ser^473 and human umbilical veine endothelial cell proliferation. Methods:To construct pAAV-CD151 and transiently transfect to HUVEC ( human umbilical veine endothelical) mediated with Lipofectamine 2000. HUVEC were assigned to control group, GFP group, CD151 group. After transfection,the expression of CD151 ,Akt and phospho-Akt were measured by western blot. Proliferation of HUVEC were measured by MTF colorimetric assay. Results :(1)The expression of CD151, phospho-Akt of pAAV-CD151 transfected group were significantly increased compared with non-transfected group and pAAV-GFP transfected group ( P 〈 0.05 ).(2) Proliferation of HUVEC of pAAV-CD151 transfected group was higher than these non-transfected and pAAV- GFP transfected group ( P 〈 0.05 ), results of which were 0.166 3 ± 0.009 5,0.169 ± 0. 008 5,0. 245 7 ± 0.0044 in MTT assay. Conclusion:We found that CD151 may phosphorylate Akt at Ser^473 and increase Akt acivity,thus induce cell proliferation. Akt is involved in CD151-induced cell proliferation.
