改进HPLC柱切换法血浆直接进样测定法莫替丁

摘　　要：建立了以RP2（25～40μm）为预处理柱，水为预处理流动相，以0．2mol／L乙酸和0．2mol／L乙酸按为净化清洗液，血浆直接进样，在线（on-line）固相浓缩净化样品；以ShimpackCLC－ODS为分析柱（15cm×6mmID），甲醇-乙酸按（0.2mol／L，pH2.7）（14：86）为分析流动相，267um波长检测，外标法定量测定人体血浆中法莫替丁的浓度的方法．该法切换系统构成简单，仪需要一个切换阀和两个溶剂系统。200μl血浆仅以。05mol／L乙酸稀释后即可进样，净化回收率为102．8％，法莫曾丁的最低检测浓度为3ng／ml。分析一个样品仅需10min。An improved method ot HPLC column switching for determination of famotidine in plasma was described. The system contained only one switching valve and two solution mobile phase, Merck ' Lichroprep RP2(25~40μm) was used as short precolumn packing material for online clean up and concentration. A Shimpack CLC-ODS (5μm) was used as ment mobile phase and a mixture of methano1~0. 2 mol/L ammonium acetate solution (14: 86) was used as anlytical mobile phase. A volume of 200 μl plasma was diluted with a solution of 0.05mol/L acetic acid and inkcted directly on the precolum- The recovery of clean up is 102. 8%. and detection limit is 3 ng/ml. Only ten minutes is needed for one sample assay.
