摘  要:目的 探讨睡眠剥夺对抑郁模型大鼠不同脑区色氨酸羟化酶(TPH)和单胺氧化酶A(MAO-A)的影响。方法 对抑郁模型大鼠进行睡眠剥夺,应用旷场实验观察大鼠实验前后自主行为的变化,用紫外分光光度计和免疫印迹技术检测大鼠大脑中脑、海马、下丘脑和前额叶四个部位的MAO-A的活性和TPH。结果 抑郁模型大鼠经过睡眠剥夺后自主行为显著增加,睡眠剥夺后大鼠海马、中脑、下丘脑和额叶的TPH显著升高,大鼠海马MAO-A活性显著增加。结论 睡眠剥夺对抑郁模型大鼠的抑郁样行为有改善作用,可能与色氨酸羟化酶和单胺氧化酶A有关。Objective To explore the effect of Rapid eye movement sleep deprivation (REMSD) on behavioral depression and activities of cerebral tryptophan hydroxylase (TPH) and monoamine oxidase A ( MAO - A) in rats treated with chronic stresses. Methods REMSD was performed by flow- erpot technique. An open field test was repeated three times for evaluating the depressive - behavior in rats during the experiment. Expression of TPH and activities of MAO - A in hippocampus, frontal cortex, hypothalamus and mid - brain were measured by western - blotting and UV - spectropho- tometer respectively. Results Open field tests showed that 72h REMSD reversed the depressive - behavior of rats remarkably. There was significant difference in activities of TPH in hippecampus, mid - brain, hypothalamus and front cortex between chronic stress rats and 72h REMSD rats. Further- more the difference in increased activity of MAO - A in hypothalamus between chronic stress rats and 72h REMSD rats was also significant. Conclusion REMSD may improve the depressive - behavior in rats. It may be correlated to the increased activities of both TPH and MAO - A .
