摘 要:对不同转TrxS基因啤酒大麦的T1,T2代植株进行分子检测,并根据检测结果对其遗传行为做了分析.结果表明:目的基因在不同材料中的遗传行为不同,TrxS基因在Harrington中符合3∶1的遗传比例,PCR-Southern杂交结果说明目的基因已整合到大麦基因组中,并能够稳定地遗传给后代;而在Franklin后代自交过程中发生丢失而不能稳定遗传.经PCR跟踪检测,得到转基因大麦纯合株系.农艺性状分析结果表明,转基因株系除生育期比对照提前4 d外,其它农艺性状与对照没有明显差异.Detection at molecular level of T1 and T2 transgenic malting barleys with TrxS gene indicated that the genetic behaviors of TrxS gene in different malting barleys were different, the segregation ratio in Harrington was consistent with Mendel' s law; PCR-Southern blot showed that the alien gene had integrated into barley genome and could be inherited to the following generations. The gene couldn' t be inherited steadily in Franklin because of being lost during self-hybridization. And analysis of several agronomic traits indicated that transgenic plants had little difference from the control except the growing period.
