早期干预对新生鼠脑神经生长相关蛋白表达及细胞凋亡的影响

摘　　要：目的探讨早期干预对正常新生大鼠神经生长相关蛋白(GAP-43)表达及细胞凋亡的影响。方法采用配对设计,30只正常新生鼠随机分为干预组与对照组,对干预组大鼠采用早期触摸14d,随后置于丰富环境14d的干预方式,对照组未予干预。通过免疫组织化学染色法和原位末端标记法(TUNEL染色)分别检测二组大鼠海马和额叶皮质GAP-43的表达情况和凋亡神经元数量。脑功能评估采用水迷宫测验。结果干预组大鼠额皮质和海马区神经元GAP-43免疫反应阳性反应较对照组显著增高(P<0.05),额皮质及海马区凋亡神经细胞中位数干预组(0;2.0个/HP)较对照组(1.0;6.5个/HP)明显减少(P<0.01)。水迷宫测试成绩干预[(5.71±1.73)次]明显优于对照组[(7.77±1.74)次](P<0.01)。结论促进GAP-43的表达和减少神经元凋亡,与早期干预促进脑发育与脑功能的机制有关。Objective To explore the effects of early interventions on growth associated protein - 43 （ GAP - 43 ） and neuron apoptosis in brain of neonatal rats. Methods According to matched - pairs design,30 rats from the same materal rats were divided into two groups.intervention group and control group randomly. The intervention group received the neonatal handling for 14 d and then were kept in an enriched environment for another 14 d. The expression of GAP - 43 and the number of neurons apoptosis were detected by immunohistochemistry and TUNEL staining respectively in forehead cortex and hippocampus of rats. The brain functional outcomes of rats were evaluated by water - maze test. Results In prefrontal cortex and hippocampus, the level of GAP - 43 immuno - positive response in intervention group was significantly higher than that of control group（ P 〈 0. 05 ）. While the median of apoptosis positive neurons in intervention group was （0 ;2.0 cells/HP） significantly lower than that of control group （ 1.0 ; 6.5 cells/HP） （ P 〈 0. 01 ）. The score of water maze test was higher in intervention group [（5.71 ±1.73）times]than that in control group[ （7.77 ±1.74）times] （P〈0. 01）. Conclusion The up -regulated GAP-43 expression and decreased the sum of neuron apoptosis may be involved in improving development and function of brain by the early intervention mechanism.
