乙肝DNA疫苗pCI/S的构建及诱导小鼠的免疫反应

摘　　要：目的:构建乙肝DNA疫苗pCI/S,观察其接种小鼠后所诱生的免疫应答效果。方法:构建重组质粒pCI/S,将其免疫BALB/c小鼠,在动态观察小鼠血清抗-HBs的诱生,并于第12周时检测体外CTL活性,同时以免疫组织化学法检测小鼠接种部位HBsAg的表达情况。结果:小鼠肌肉接种pCI/S后,1周时血清抗-HBs开始阳转,阳转率于6周、8周达高峰,且能诱生特异性CTL应答。此外,免疫组织化学法能在小鼠接种局部检测出HBsAg的表达。结论:乙肝DNA疫苗pCI/S能在小鼠体内诱生特异性的体液免疫和细胞免疫应答。Objective： To constructed HBV DNA vaccine pCI/S, observe specific humoral and cellular immune responses in BALB/e mice being inoculated with pCI/S. Methods： Recombinant plasmid pCI/S was constructed and was intramusclarly injected into BALB/e mice, and boosted after 1 and 2 month. The production of the expressed protein in muscle tissues in situ and specific eeUular immune responses were detected 3 months after the first injection, specific humoral immune responses were detected at 1, 2, 4, 6, 8, 12week after the first injection. Results： After intramusclar injection of pCI/S into BALB/e mice, HBSAb were elicited and eytotoxie T lymphocyte responses were induced and HBsAg protein was expressed in situ in muscle tissues. Conelusions： pCI/S is effective HBV DNA vaccine that can induced specific humoral immune responses and eeUular immune responses.
