摘  要:目的 研制一种简便快速检测鼠伤寒沙门菌的新方法。方法 应用胶体金技术和免疫层析原理进行研究,建立检测沙门菌的胶体金免疫层析技术(GICA),并研究GICA检测鼠伤寒沙门菌的敏感性、特异性和稳定性。结果 每毫升胶体金标记兔抗鼠伤寒沙门菌IgG的最适量为24μg,最佳抗体包被浓度为1mg·ml^-1。Millipore公司0.45μm的硝酸纤维素膜蛋白吸附性能最好。建立的GICA检测试条检出鼠伤寒沙门菌的下限为10^5cfu·ml^-1,达到检测技术指标。从检测到结果判断时间仅5~10min。GICA检测试条分别在4C和室温下保存半年,检测结果不变。结论 应用GICA检测鼠伤寒沙门菌的方法具有快速、简便、敏感、犍异和稳定的特点,非常适合基层卫生单位常规检测应用。Objective To develope a rapid and simple new method to detect Salmonella typhimurium. Methods Gold immunochromatographic assay(GICA) was established to detecting Salmonella typhimuriurn by combing colloidal gold technique with immunochromatographic assay. Besides its sensitivity,specificity and stability were studied. Results The optimum marked amount of IgG(rabbit anti mouse) of one-milliliter colloidal gold was 24μg, the optimum concentration of the coating Ab was 1mg· ml^-1. The NC film with the best absorbing capability for protein was selected from Millipore company,which diameter was 0.45μm. The prescribed minimum of GICA test strip to detect Salmonella typhimurium was 10Scfu·ml^-1 ,which attained the techenical index. It took only 5-10 min from detecting to judging the result. GICA test strip could be stored at 4C and room temperature for half a year,which showed the same result. Conclusion GICA detecting Salmonella typhimurium is rapid,sensitive,specific and stable. It is particularly fit for routine test of country sanitary unit.
