摘  要:用聚丙烯酰胺凝胶电泳(PAGE)法,分离了不同生育期菊芋叶片过氧化物酶(Helianthus tuberosus.1eves peroxidase,HLP)同工酶,并测定了HLP的酶活性;比较了菊芋叶及其他4种植物POD的特点。结果表明:(1)同一植株不同部位的HLP酶活性大小顺序为,中部成熟叶(6.89×103U/g)>底部衰老叶(5.38×103U/g)>上部嫩叶(2.82×103U/g);由嫩叶→成熟叶→衰老叶的发育过程中,HLP同工酶酶带共有9条,出现的方式有稳定型(Rf 0.81带)、酶活性先弱后强型(Rf0.52、0.67、0.74、0.85带)、酶带先有后无型(Rf 0.70、0.72带)、酶带先无后有型(Rf 0.77)4种。(2)几种植物POD的酶活性的大小顺序是:甘蔗叶>竹笋壳>菊芋叶>大豆壳;POD同工酶电泳最大迁移率(Rf)大小顺序是:菊芋叶(0.85)>甘蔗叶(0.70)>竹笋壳(0.61)>大豆壳(0.57)>辣根(0.55),HLP同工酶属阴离子型。探讨了菊芋POD的表达特点。Peroxidase isoenzymes of jerusalem artichoke (Helianthus tuberosus. ) leaves in various growth periods were seperated by means of PAGE. Peroxidase enzyme activities of jerusalem artichoke leaves in various periods were determined and compared with POD of other 4 plants. It was found that POD enzyme activities in different parts of the plant were ordered as follow: mature leaves in the mid part (6.89× 10^3U/g) 〉 old leaves in the lower part (5.38× 10^3U/g) 〉 new leaves in the upper part (2. 82 × 10^3U/g) ;9 POD isoenzymes bands appeared in the growth process from new leaves, mature leaves to old leaves. Four kinds of appearances were included: band of Rf 0.81 was stable; isoenzymes activities of bands of Rf 0. 52, 0.67, 0.74, 0.85 first weak then strong; band of Rf 0.70, 0.72 appeared first then disappeared; band of Rf 0. 77 disappeared first then appeared. The POD enzymes activities were ordered as follow, cane leaves 〉 bamboo shell 〉 jerusalem artichoke leave 〉 soybean hull; Rf values of POD isoenzymes were as followed: jerusalem artichoke leave (0. 85) 〉 cane leave (0.70) 〉 bamboo shell (0.61) 〉 soybean hull (0.57) 〉 horseradish (0. 55). jerusalem artichoke peroxidase isoenzyme is of negative ion. The characteristics of jerusalem artichoke POD - isoenzymes expression were also discussed.
