海南菜豆上粉虱传双生病毒的分子检测

摘  要:为了解粉虱传双生病毒在海南的发生情况,从海南儋州采集了3个菜豆病样,症状表现为花叶、皱缩、叶背部叶脉突起,根据粉虱传双生病毒基因组DNA基因间隔区及外壳蛋白基因保守序列设计的简并引物,对这3个样品进行了PCR检测,从其中两个样品中扩增到预期大小为522 bp的DNA片段。PCR产物经克隆后进行序列测定,BLAST分析结果表明,该序列与与印度绿豆黄花叶病毒豇豆分离物(GenBank登录号:AF481865)的同源性最高,为65.9%,说明海南菜豆中存在着粉虱传双生病毒的侵染。To investigate the occurrence of whitefly-transmitted geminivirus in Phaseolus vulgaris L. in Hainan,three virus-infected Phaseolus vulgaris L. samples with mosaic, crinkle and vein plum symptom were collected in Danzhou, Hainan Province. A degenerate primer pair was designed according to the conserved sequence of CP gene and intergenic spacer of WTG,and a single PCR product of expected size (522 bp) was amplified from two samples by PCR, cloned and sequenced. BLAST analysis showed that PCR product had a geminiviral gene origin,and the sequence had the highest sequence identity (65.9%) with that of Mungbean yellow mosaic India virus-[Cowpea] (AF481865), which indicated that two samples were infected with geminiviruses.
