槲皮素对LPS刺激肺上皮细胞核转录因子NF-κB表达的影响

摘　　要：目的研究槲皮素对肺上皮细胞株(A549细胞)不同时段表达核转录因子(NF-κB)的影响。方法用LPS建立体外A549细胞损伤模型,用免疫印迹法(Western Blot)观察不同时间A549细胞核转录因子NF-κB的表达。结果A549细胞在LPS刺激下,NF-κB表达水平逐渐升高,呈现出一定的时间依赖性,而槲皮素能显著抑制其表达。结论NF-κB是炎症反应中重要的核转录蛋白,参与一系列炎症介质的表达。因此,槲皮素抑制LPS诱导A549细胞产生NF-κB,提示槲皮素可能通过对炎症因子负性调控而发挥其抗炎作用。Objective To study the influence of Quercetin on the expressions of NF-κB in pulmonary epithelial cell line（A549） in different periods.Methods The experiments were performed in culture of LPS induced A549 in vitro,Tne expression of NF-κB was detected with western blotting. Results A549 cell was stimulated with LPS and the activaty of NF-κB was increased time dependence, but it was inhibited by Quercetin. Conclusion Quereefin might exerts anti-inflammatory effects through negatively modulating pro-inflammatory factors.The inhibitory effects of quereetin on NF-κB production by A549 cell may provide a theoretic basis on future therapy of inflammation.
