均衡的脱氧核苷增强^(125)I-UdR杀伤效应的实验研究

摘  要:目的研究均衡的脱氧核苷混合物(AUCG)对5-^(125)I-2′-脱氧尿嘧啶核苷(^(125)I-UdR)杀伤效应的增强作用及机制。方法在鼠 C6神经胶质瘤细胞培养液中加入^(125)I-UdR(74 kBq/ml)及不同浓度的 AUCG,依据细胞集落形成实验结果,计算在 AUCG 不同浓度和不同作用时间下的细胞存活分数(SF),并通过细胞形态学变化、透射电镜(TEM)及 DNA 凝胶电泳观察 AUCG 增强作用的结果。结果不同 AUCG 浓度的各实验组 SF 均显著低于对照组(P<0.01),且随着 AUCG 浓度的增加,各实验组之间的 SF 逐渐下降。与低浓度组(AUCG≤20 μmol/L)比较,各高浓度组(AUCG≥50 μmol/L)的细胞存活水平明显降低(P<0.01)。同时 AUCG 对^(125)I-UdR 的增强作用与时间呈正相关,SF 随着AUCG(30 μmol/L)的作用时间延长而逐渐减少。通过 TEM 可观察到细胞凋亡小体形成,凝胶电泳结果呈现 DNA 梯度条带,提示细胞出现明显凋亡。结论 AUCG 能够增强^(125)I-UdR 对 C6细胞的杀伤效应,并促进细胞凋亡的增加。Objective A potential anti-tumor agent, 5-125^I-Iodo-2′-deoxyuridine( UdR), was cellular cycle dependent. The experiment was thus designed trying to enhance the lethal effect of UdR by a balance deoxyribonucleoside mixture (AUCG). Methods After adding into medium of rat C6 cells with the mixture of 125^I-UdR (74 kBq/ml) and AUCG at different concentrations, the survival fractions (SF) were determined using plating efficiency assay, The enhancement, meanwhile, was observed through transmission electron microscope (TEM) and DNA agarose gel electrophoresis. Results Compared to control, the SF decreased in cells treated with AUCG (P 〈 0.01 ), The decrease of SF was positively related to AUCG concentrations, namely, the higher dose( ≥50 μmol/L) produced more evident SF reduction ( P 〈 0.01 ), The enhancement was found increased with duration of treatment, Apoptotic bodies was observed through TEM and DNA ladder strip obviously appeared on agarose gel electrophoresis, suggesting the noticeable apoptosis in the group cultured with 125^I-UdR (74 kBq/ml) and AUCG (30 μmol/L), Conclusion A UCG might enhance the lethal effect of 125^ I-UdR through apoptosis of C6 cells,
