摘 要:目的观察金线莲提取物对小鼠急性和慢性化学性肝损伤的保护作用。方法分别应用水煎法、醇提法、全草榨汁法提取金线莲中的有效成分;小鼠分别腹腔注射0.1%CCl4的蓖麻油溶液、灌胃50%CCl4的蓖麻油溶液5周,制作急慢性化学性肝损伤模型;分别将金线莲3种提取物以1.0、5.0g.kg-1给小鼠灌胃,以生理氯化钠溶液和益肝灵片(主要成分为水飞蓟素)分别作空白和阳性药对照;通过小鼠血清生化指标(谷丙转氨酶和谷草转氨酶)测定、脏器称重和肝脏病理切片观察,评价金线莲各提取物对小鼠急、慢性化学性肝损伤的保护作用。结果金线莲提取物可使CCl4所致急、慢性化学性肝损伤小鼠血清中的谷丙转氨酶和谷草转氨酶降低50%以上(P<0.01),转氨酶值接近正常组;金线莲提取物可抑制肝损伤引起的肝脏和脾脏肿大以及胸腺萎缩;肝组织切片镜检,金线莲提取物能明显减轻中毒小鼠的肝损伤以及早期肝纤维化的形成。结论金线莲对小鼠急、慢性化学性肝损伤具有保护作用及显著的降酶作用,并能减少早期肝纤维化的发生。Aim To evaluate the effect of Herba Anoectochili extracts on acutely and chronically damaged livers induced by CCl4 in mice. Methods The damaged liver models were established by ip 0.1% for a single injection or by ig 50% CCl4 in mice once every five days for 5 weeks. Then they were treated with Herba Anoectochili extracts or saline. The liver, spleen, thymus and blood of mice were taken. The weight of immue organs, the pathological changes in the liver and activities of alanine transferases (ALT) and aspartate transferanses (AST) in serum were determined. Results Pathological changes of liver induced by CCl4 in mice treated with Herba Anoectochili extracts were not more significant than those treated with saline. The levels of ALT and AST in the serum of mice administered by Herba Anoectochili extracts were decreased by more than 50% compared with those administered by saline.
