摘 要:目的:优化黄芩苷提取分离工艺。方法:采用正交设计,利用高效液相色谱法(High performance Liquid Chromatography,HPLC)检测生药粒度、不同提取纯化方法对黄芩苷纯度及得率的影响,优选提取纯化方案。以优选方案为实验组,常规煎煮法为对照组,进行平行实验。结果:20目生药粒度,微波提取6 min,超滤柱过滤后酸沉为优选的提取方案,黄芩苷得率为5.77 g%,纯度为89.98%。常规煎煮法黄芩苷得率为4.55 g%,纯度为77.77%。平行实验结果表明,优选方案在黄芩苷纯度及得率上都要明显高于常规煎煮法(P<0.05)。结论:微波提取、超滤柱过滤后酸沉的提取方法能够改善黄芩苷的提取率和纯度。Objective:To optimize the extraction and purification methods of baicalin from radix scutellariae. Methods:As compared to the conventional decoction method, orthogonal analysis was applied to new extraction and purification methods of baicalin by comparing the purity and yield rates of baicalin. A high performance liquid chromatography assay was used for measurements. Results:After a 20- mesh in granular particle size of radix scutellariae in water solution was treated with microwave extraction for 6 min, filtered by the ultra-filtration column and precipitated by acid, the yield rate of baicalin was 5.77% and purity of radix scutellariae was 89.98%. In contrast, the yield rates and purity were 4.55% and 77.77% obtained by conventional methods, exhibiting evident improvements on the extraction and purification methods ( P 〈 0.05 ). Conclusions : The microwave treatment and uhra-filtration followed by acid precipitation methods could improve the extraction and purification of baicalin from radix scutellariae.
