摘  要:自20世纪90年代DNA疫苗在免疫学上被第一次提出来以后,10多年来DNA疫苗以其独特的优点而倍受青睐,但近来越来越多关于其免疫效力低下和对灵长类及人等大型动物收效甚微的论证制约着其广泛的应用。即是从DNA疫苗免疫机制方面对构建优化疫苗载体的最新研究进展综述。DNA vaccine was put forward for the first time in the nineties of the twentieth century,it has been admiring for its special advances over decades years.At that time it appeared to be a revolutionary new approach in vaccinology.Since then,genetic immunization has been applied with much success in a wide variety of model and natural systems.It has also been used in several human clinical trials.However,.the technique is currently thought to be poor at antibody production and more importantly not to work well in primates and humans (simian barrier).Many more experiments have been carded out to evaluate different factors that determine the efficiency of gene transfer and immunogenicity of plasmid DNA.In this review we discuss lots of strategies up to date of constructing effective vaccine plasmids in the base of immunity mechanism.
