四季桔试管苗基因组DNA微量提取方法

摘　　要：本试验以四季桔试管苗为试材,进行基因组DNA微量、快速提取方法的研究。结果表明:采用改良CTAB法,仅需20mg样品、2.5h就能获得高纯度、高质量的DNA,这是目前木本植物样品量最小的DNA提取方法;提取的DNA能直接被限制性内切酶BamHⅠ酶切,也能进行PCR扩增,可用于进一步的分子生物学研究。A revised method of CTAB for trace and rapid extraction of genomic DNA from the tube pladets was developed in Citrus microcarpa Bonge.The experimental results showed that the high-quality and high-purity DNA was obtained by the revised CTAB method from only 0.02g sample for only 2 hours,which was the minimum of the sample for genomic DNA extraction in woody plants;and the extracted DNA could be digested by BamH Ⅰ and used for the PCR analysis for further studies on molecular biology.
