重组人干扰素-γ对角朊细胞体外增殖分化影响的研究

摘　　要：以人转化型角朊细胞株 Co L o- 16细胞为模型 ,通过活细胞计数、3H- Td R掺入法、电镜及免疫组化方法研究了重组人干扰素 -γ( rh IFN-γ)对角朊细胞体外增殖分化的影响。结果表明 IFN-γ能以一种剂量依赖方式抑制人角朊细胞株 Co L o- 16细胞的体外增殖和 DNA合成 ,对角朊细胞核有直接损伤作用 ,并抑制角朊细胞角蛋白表达。提示The culture of human transformed keratinocyte cell line Co Lo- 16cells were used as a model,and the effects of recombinant human interferon- γ( ifn- γ) on the proliferation and DNA synthesis were ob- served as determined by viable cell count,3H- Td R uptake,electronic microscopy,and immunohis- tochemistry.The inhibitory effects were dose- dependent.The nuclear of keratinocytes was directly dam- aged and the keratin expression was suppressed.This suggests that IFN -γ may change keratinocyte be- havior of proliferation and differentiation in vitro.
