摘 要:目的观察血红素氧合酶-1(HO-1)的诱导剂正铁血红素(hemin)对胰岛素抵抗(IR)大鼠胸主动脉的舒张功能及形态的影响,并对其机制进行初步探讨。方法通过6周高脂饮食成功复制胰岛素抵抗SD大鼠模型,分为使用正铁血红素(hemin组)与锌原卟啉(ZnPP组)对IR大鼠进行腹腔注射共4周,观察收缩压、血一氧化碳含量、胸主动脉氧化应激指标、胸主动脉舒张功能以及胸主动脉内皮扫描电镜下的改变。结果Hemin可升高IR大鼠胸主动脉组织的总抗氧化能力(TAOC)及超氧化物歧化酶(SOD)水平,降低肿瘤坏死因子(TNF)-α水平,提高胸主动脉血管环对乙酰胆碱的舒张反应,改善胸主动脉内皮的形态学损害。结论HO-1的诱导剂hemin通过抗氧化、抗炎及调节血管活性物质等方面参与改善IR大鼠血管病变,对早期IR大鼠血管功能起保护作用。Objective To investigate the effects of heroin (inducer of HO-1)-on diastolic function and morphology in the thoracic aortas of insulin resistance (IR) rats and demonstrate the probable mechanisms. Methods IR model of Sprague- Dawley (SD) rats was successfully established by feeding with high fat diet (HFD) for 6 weeks. Then the model rats were randomly divided into 2 subgroups, namely, hemin group and zinc protoporphyrin-IX (ZnPP-IX) group, and were treated every other day with abdominal injections of hemin or ZnPP-IX for 4 weeks. Systolic arterial blood pressure (SABP), blood carbon momxide (CO), oxidative stress markers in thoracic aorta, diastolic function of thoracic aorta, and the ultrastructure of thoracic aortic endothelium was assessed. Results Heroin increased the levels of total antioxidant capacity (TAOC) and superoxide dislnutase (SOD), decreased the level of tumor necrosis factor (TNF)-α, improved the vasorelaxation of thoracic aortic rings (TARs) to ACh, and attenuated structural damages of rat thoracic aortic endothelium. Conclusions These studies suggest that the protective effect of hemin (inducer of HO-1) against vascular injury in IR states was due to the improvement of vasculopathy by anti-oxidation, anti-inflammatory effects and regulation of vasoactive suhstance.
