单次静脉注射丙泊酚的催眠量效关系

摘　　要：目的研究单次静脉注射丙泊酚的催眠量效关系。方法将30例ASAⅠ～Ⅱ级全麻病人分为6组,每组5例,分别给予丙泊酚0.8、1.0、1.2、1.3、1.5和1.8mg/kg静脉注射;以睫毛反射消失,大声呼唤无睁眼为入睡标准。采用probit法和logit法计算丙泊酚的半数有效剂量(ED50)及95%有效剂量(ED95)。结果probit法丙泊酚的催眠ED50值(95%可信限)为1.14(0.93,1.35)mg.kg-1,ED95值(95%可信限)为1.66(1.38,3.12)mg.kg-1;logit法丙泊酚的催眠ED50值(95%可信限)为1.14(0.96,1.37)mg.kg-1,ED95值(95%可信限)为1.67(1.38,4.06)mg.kg-1。结论probit法丙泊酚的催眠ED50值(95%可信限)和ED95值(95%可信限)分别为1.14(0.93,1.35)mg.kg-1和1.66(1.38,3.12)mg.kg-1。Objective To define the median and 95% effective dose of hypnotic of propofol. Methods Thirty patients, ASA grade I- II, non premedical patients undergonging elective surgery were random divided into 6 groups. The patients of Each groups received 0.8,1.0,1.2,1.5,1.8 or 2.2 mg.kg^-1 of propofol intravenous bolus respectively. The means values of 50% patients effective dose （EDso） and ED95 were obtained by probit analysis and logit analysis at the endpoint of hypnotic （loss of response to verbal command and loss of eyelash reflex）. Results By probit analysis, the EDs0 and EDgs values of propofol at hypnotic （95% confidence interval） were 1.14（0.93, 1.35） mg.kg^-1 and 1.66（1.38, 3.12）mg.kg^-1 respectively. By logit analysis, the ED50 and ED95 values of propofol at hypnotic were 1.14 （0.96, 1.37） and 1.67 （1.38, 4.06） mg.kg^-1 respectively. Conclusions By probit analysis, the EDso and EDgs values of propofol at hypnotic （95% confidence interval） were 1.14（0.93, 1.35） mg.kg^-1 and 1.66（1.38, 3.12） mg.kg^-1 respectively.
