摘 要:目的探讨不同类型癫痫发作对大鼠认知功能的影响。方法戊四氮点燃大鼠癫痫持续状态(SE)、慢性癫痫(CEP)和非惊厥性癫痫持续状态(GNCSE)三种癫痫动物模型,采用交替电刺激Y迷宫、抬高迷宫和旷场试验观察致痫后不同时间点大鼠学习记忆功能和情感反应变化。结果SE大鼠致痫后1d电Y迷宫错误次数TE[(10.0±3.0)次]多于对照组[(7.7±2.7)次];抬高迷宫开放臂中逃避时间[(57.4±34.0)s]长于对照组[(34.2±15.2)s],进入次数[(3.5±1.7)次]多于对照组[(2.2±1.1)次];旷场活动的格子数(44.8±31.4)格、站立次数(5.6±3.3)次及排出粪便颗粒数(1.3±1.7)粒均少于对照组[分别为(78.2±42.7)格,(9.3±5.0)次,(3.1±2.4)粒]差异具有显著性(P〈0.05)。而30d均无差别。CEP大鼠电Y迷宫TE在1d和30d均增多(P〈0.05);抬高迷宫开放臂中停留时间及进入次数和旷场活动格子数、站立次数及粪便颗粒数与对照组相比均无明显差别(P〉0.05)。GNCSE组大鼠1d电Y迷宫TE有所增加,但差异不显著(P〉0.05),30d差异无显著性;且抬高迷宫和旷场试验与对照组相比亦无明显差异(P〉0.05)。结论癫痫发作可以导致实验动物活动习性改变、警觉水平降低、情感行为异常及学习和记忆能力下降等多种认知功能受损的表现,而且不同类型的癫痫发作对认知功能的影响有所不同。Objective To explore the effect of different types of the epileptic seizures on the cognition of rats, searching for the most apparente epileptic animal model which was affected for the cognition. Methods Spraque-Dawley(SD) rats were used to establish the different types of experimental epilepsy models kindled by PTZ. The function of learning and memory in different types of epileptic model rats was examined by the alternative electro-stimulus Y-maze test(AESYT). The change of emotional behavior in different types of epileptic model rats was examined by the elevated plus maze test(EPMT) and open field test(OFT) at 1 day and 30 day after epilepsy. Results The results of AESYT in status epilepticus (SE) rats showed that the total error (TE) were higher than corresponding control rats at 1 day after epilepsy(P〈0.05) , and there were no different in TE at 30 day after epilepsy( P 〉0.05). In SE rats, the results showed that it was increased in the times of visits as well as the time spent on the open arms of EPMT ( P 〈 0.05 ) at 1 day after epilepsy, and there were no different at 30 day comparing with corresponding control rats ( P〉 0.05 ). The results of OFT in SE rats showed that it was decreased in the number of squares crossed, rearings and fecal boll ( P〈 0.05 ) at 1 day after epilepsy, there were no different at 30 day comparing with corresponding control rats(P〉0.05). It was showed that the TE in AESYT of chronic epilepsy (CEP) rats were higher than corresponding control rats at 1 day and 30 day after epilepsy( P 〈 0.05 ) , but there were no different at any time comparing with corresponding control rats in EPMT(P〉0.05) and OFT( P 〈0.05 ). In generalized nonconvulsive status epilepticus (GNCSE) group , the TE in AESYT were not higher than control rats weather at 1 day or 30 day after epilepsy(P〉0.05).
